When I clean Spyware, I boot to safemode and run HJT. I disable all the entries that are either spyware or not necessary (look up unknown entries on castlecops.com). This prevents the spyware from starting with the computer, which is what causes the symptoms: pop-ups, slowdown, memory usage, etc. Actually removing the executable files of Spyware isn't a huge deal in my opinion. However, if you are so inclined, Dr. Web's Cure-it does a great job for spyware/trojans/viruses, and it is free. I also second the person who mentioned ComboFix. Running Spybot S&D and those programs can't hurt.
Microsoft Anti-spyware (aka Windows Defender) provides good real-time protection in Vista (and XP if you download it). Also, I'm a huge fan of Spyware Guard and Spyware Blaster from javacool in terms of preventing future infection. Having up-to-date Firefox, Adobe Reader, Adobe Flash Player, Shockwave, and Sun Java Runtime is also highly important. And of course Windows update (which now includes IE7, which is very important... even if the user uses Firefox, some existing Spyware launched IE in the background; having the more secure IE7 can prevent further infection).
Spybot Search and Destroy, and Ad-aware combination works best for me.

You should have Ad-Aware (Classic), Spybot Seek & Destroy & Spywareblaster in your tool kit. You might want to include HiJackThis & Process Explorer (http://www.microsoft.com/technet/sys...s/default.mspx).

For a computer technician or according to a computer technician?
For a computer technician, I would say a Registry Editor, the Windows Command line and BartPE. There are no anti spyware more efficient that those tools in the hand of a computer technician.
Otherwise, I think Ad-Aware and spybot S&D work reasonnably well. Do the scans in Safe-Mode, you have more chances of removing spywares.
