More than 1,000 people are expected to take part in the 10km event in east Manchester on July 13 and competitors are still being sought.
The Urbanathlon, the first of its kind in Europe, has been launched by Manchester Olympian Diane Modahl at Sportcity.
Racers will begin with a lap of the regional athletics track, before racing around the B of the Bang sculpture and up and down the stairwells of the City of Manchester Stadium.
They will then tackle a series of 13 unusual obstacles on Joe Mercer Way and around Clayton Vale, including fording the River Medlock, before the finish the 10km race in Philips Park.
Diane said: "There's nothing quite like this in Europe. It's a great opportunity for anyone who loves sport to get involved and test themselves - when else would you get the chance to scramble over cars and over all sorts of different obstacles?
"This is great for east Manchester as it continues the fantastic legacy that was left behind after the Commonwealth Games. Clayton Vale is a beautiful part of Manchester, but it's really hidden away."
The urban footrace is open to anyone older than 16 and competitors can run alone or as part of a team. If successful, it will become an annual event.
It is being organised by Sporting Enterprises and Sport 4 Life, and is supported by Manchester Council and regeneration body New East Manchester to showcase the area.

It was the "Delicate Sound of Thunder" tour - they did stuff from Meddle (One of those Days), Dark Side of the Moon, Animals, Wish You Were and some from the Wall and Momentary Lapse of Reason. It was not long after Roger Waters left. It was a cracking gig - lovely, still, warm, summers evening.

I wish I had seen them before Roger left the band. He was the heart and
brains of Floyd. However, I'll give David his due. His guitar playing was
the soul of the band. His guitar playing on Wish you were here is the
best, recorded acoustic I have heard to date (Rock).
